The 25-year-old replaces veteran Danish defender Michael Jakobsen, who held the role last season.
"This isn't something I necessarily expected but I'm honoured," Mauk said on Tuesday.
"I'm not sure if the gravity of this has truly sunk in yet but I understand how privileged I am to be given this responsibility."
Mauk, who first joined Adelaide midway through the 2015-16 season and returned in January this year after a spell abroad and stints with Melbourne City and Brisbane, was described as a "natural leader" by coach Carl Veart.
"In Stefan we have someone who strives for team excellence every day and is someone who I believe has the necessary respect of the entire football club to drive us further forward," Veart said.
